Comparison of UV spectrophotometry and HPLC on quantitative determination of total flavonoids of Herba Epimedii

Abstract

To investigate the quantitative determination of total flavonoids from Herba Epimedii using UV spectrophotometry and HPLC.Methods:Icariin was used as the reference compound for UV spectrophotometry,determined at 270 nm.HPLC was performed on a ZORBAX SB-C18 analytical column(250 mm×4.6 mm,5 μm)with gradient elution of acetonitrile and water at the flow rate of 1.0 mL·min-1.The UV detection was set at 270 nm.The peaks with similar UV spectra to that of icariin were identified as flavonoids, and ten of them were confirmed by ESI-MS.The contents of epimedin C,icariin,2”-O-rhamnosylicariside and baohuoside were quantitated with their individual calibration curves,respectively.The content of other flavonoids was estimated using icariin as the reference compound.The content of total flavonoids from Herba Epimedii was the sum of epimedin C,icariin,2”-O-rhamnosylicariside , baohuoside and other flavonoids.Results:The linear range of icariin was 2.45-24.50 μg·mL-1 for UV spectrophotometry(r=0.9996).The content of total flavonoids from Herba Epimedii calculated with reference to icariin was 56.6%.For HPLC analysis,epimedin C,icariin,2’’-O-rhamnosylicariside and baohuoside had good linearity in the ranges of 0.093-1.852 μg(r=0.9998),0.107-2.136 μg(r=0.9997),0.094-1.876 μg(r=0.9998),0.098-1.956 μg(r=0.9998),respectively.The content of total flavonoids from Herba Epimedii was 35.6%.Conclusion:The contents of total flavonoids from Herba Epimedii determined by UV spectrophotometry and HPLC are significantly different, which suggests that the accuracy of UV spectrophotometry is suspectable.
